Cyclic stretch inhibits LPS/ATP-induced activation of caspase-1. J774.1 cells were exposed to cyclic stretch of 20% elongation at a frequency of 10 cycles/min for the first 2 h during treatment with 100 ng/mL LPS for 4 h followed by stimulation with ATP for 2 h in the continuous presence of LPS. a Concentrated supernatants were analyzed by western blotting with specific antibodies to caspase-1 and IL-1β. b Cells were labeled with a FLICA probe conjugated with FAM (green) and nuclei were visualized by staining with Hoechst 33342 (blue) (magnification, × 200; scale bars are 50 μm).
